Trypsin Inhibitors And Why Legumes Need Cooking
Raw legumes contain proteins that block the enzymes digesting protein, which is one reason beans are cooked thoroughly and why heat treatment improves their protein availability.

Legumes are never eaten raw, and the usual explanation stops at digestibility. The more specific reason involves proteins that interfere with digestion directly.
The plant's own defense
Seeds represent a plant's investment in reproduction, so they carry chemical defenses against being eaten. Several of these target the digestive machinery of the animal eating them.
Protease inhibitors are among the most consequential. They bind to digestive enzymes such as trypsin and chymotrypsin and block their active sites.
With those enzymes disabled, dietary protein is broken down less completely, so fewer amino acids are absorbed from the same amount of food.
A secondary effect on the pancreas
When digestion is incomplete, the pancreas responds by secreting more enzymes. Those enzymes are themselves proteins built from amino acids.
Sustained high intake of active inhibitors therefore costs the body amino acids in enzyme production while simultaneously reducing amino acid uptake from food.
This is a documented effect at high inhibitor levels rather than a concern with normally prepared legumes, where heat has already removed most of the activity.
Heat is the reliable deactivator
Protease inhibitors are proteins, which means sufficient heat unfolds them and destroys their ability to bind enzymes.
Wet heat works far better than dry heat, because water assists the unfolding. Boiling and pressure cooking are considerably more effective than roasting.
Time and temperature both matter, which is why undercooked beans remain a problem and why soaking followed by a full boil is the standard preparation.
Lectins are a related but separate issue
Lectins are proteins that bind carbohydrate structures, including those on intestinal cells. Certain beans, notably raw kidney beans, contain a form capable of causing acute illness.
Food safety guidance is specific about this: soaking followed by a vigorous boil deactivates it, while low-temperature cooking such as an unheated slow cooker cycle may not.
Canned beans have already been heat processed, which is why they present no such concern and require only reheating.
What this means for preparation
Soaking, discarding the soak water and boiling thoroughly addresses inhibitors, lectins and some of the oligosaccharides associated with digestive discomfort at once.
Fermentation and sprouting reduce inhibitor activity as well, which is part of why tempeh and sprouted legume preparations digest differently from the raw seed.
The practical consequence is that measured protein digestibility for legumes reflects cooked legumes. Preparation is part of the nutritional value, not separate from it.
